Abstract

Ankyrons® are next-generation, recombinant binding reagents designed to overcome key limitations of conventional research antibodies. These ~15 kDa single-domain binders are based on an ankyrin-repeat scaffold and selected entirely in vitro without animal immunisation. This high-throughput screening system enables rapid generation of multiple high-affinity clones against protein targets which are expressed as part of the workflow. The current Ankyron catalogue comprises over 20,000 distinct clones targeting more than 4,000 proteins from over 150 species. This includes proteins from key pathogens and microorganisms, alongside species-specific immunology targets and defined cell-type markers. The platform also supports researcher-led requests for Ankyrons against new protein targets. We demonstrate how the small size of Ankyrons, absence of an Fc region, and flexible labelling formats provide advantages for research applications including flow cytometry, immunofluorescence, and immunohistochemistry. Ankyrons offer accessible and consistent binding reagents for reliable target detection.
